Covaris, OmniSeq, and Dynamic Devices

OmniSeq, Inc. uses Covaris AFA® (Adaptive Focused Acoustics®) as an essential technology in their molecular and immune profiling of patient tumors.  Covaris AFA enables significantly higher extraction yields of DNA and RNA from FFPE samples allowing more patients to receive invaluable data from these impactful tests.  Due to the success of this extraction workflow and genetic testing, Omniseq’s throughput increased to a point where automated sample extraction is required.  Covaris and Dynamic Devices collaborated to successfully build an automated extraction platform using the integration friendly Covaris LE220R-plus Focused-ultrasonicator and Dynamic Devices Lynx LM1200 Liquid Handler.

Covaris and Ginkgo Bioworks

Ginkgo Bioworks is optimizing Covaris AFA (Adaptive Focused Acoustics) to enable their DNA and RNA extractions in a high throughput, truly automatable workflow.  Gentle and efficient lysis of microorganisms is essential to achieve quality nucleic acid extraction.  Conventional cell lysis mechanisms such as bead beating, chemical, or enzymatic treatments are time-consuming and not truly amenable to automation.  Nucleic acid yield and quality can be highly variable.  High molecular weight DNA is available for Illumina or long read sequencing library prep.  RNA is simultaneously extracted and sheared by AFA in a dose-dependent manner, ready for downstream Illumina library prep.

A novel solution to FFPE extraction challenges for downstream clinical assays

Extracting nucleic acids from FFPE samples for downstream clinical assays including Next-Generation Sequencing (NGS) is a labor-intensive process that is highly variable depending on the sample and extraction method. Covaris has developed a novel solution to address these extraction challenges. Using Adaptive Focused Acoustics® (AFA®), Covaris truXTRAC FFPE enables high throughput, reproducible extraction of high yield, NGS-grade nucleic acids from FFPE, reducing QNS rates. This session explores how truXTRAC FFPE enabled OmniSeq, Inc., a CAP accredited molecular diagnostic laboratory, to improve overall comprehensive molecular and immune profiling of patient tumors using a novel process for automated DNA and RNA extraction.
