The Covaris Adaptive Focused Acoustics® (AFA®) process employs focused bursts of very high frequency, and in some applications, very intense mechanical energy, transmitted acoustically through a water bath to a small focal zone within a sample vessel immersed in the bath.
The sample tube is a critical component and link in the acoustic wavepacket pathway. For example, the AFA-TUBE®, microTUBE, and miniTUBE were engineered by Covaris to provide controlled isothermal and repeatable treatment at all power settings.

The Covaris AFA-TUBE TPX is a novel polymer vessel designed and engineered for Focused-ultrasonicators utilizing AFA technology.
The AFA-TUBE TPX is made of a unique polymer for low impedance and better transmission of acoustic energy. The material focuses and efficiently couples acoustic energy and optimizes cavitation, resulting in the tight size distribution of DNA fragments required for NGS workflows that are necessary for NGS workflows. This unique configuration generates uniformly distributed cavitation facilitating an unprecedented standardized sample processing, which in turn increases the efficiency and reproducibility of nucleic acid shearing. The new plate format also allows for use as typical labware with compatibility with heat blocks, magnets, and thermocyclers. It can easily be used on an automation platform in an NGS workflow improving turnaround time and throughput. With no transfer steps necessary, a complete library prep can be run in AFA-TUBE TPX reducing cost of consumables and any sample loss.
The Covaris microTUBE with an AFA fiber is used extensively for DNA shearing by major genome centers throughout the world as the “best practices” for generating of deep coverage, unbiased fragments for advanced sequence analysis although they are also used for other applications with a total volume less than 130µl (e.g., FFPE thin microtome section samples). The microTUBE was specifically designed to optimize inertial cavitation and focus acoustic energy in small sample volumes (e.g., 50 microliters in the screw-cap microTUBE). The integrated AFA fiber provides nucleation sites and allows for both uniform and faster sample processing. The microTUBE is made with a special grade of hardened glass for low acoustic impedance and better transmission of energy without the generation of residual heat. The extreme AFA turbulence readily transfers excess heat from the sample to the surrounding water bath. Indeed, samples processed with the Covaris AFA system are noted for high recovery high precision and the microTUBE geometry provides synergistic acoustic focusing.

Covaris 96 microTUBE plates and strips are designed for the automated batch processing capability of the E-Series and higher throughput, L-Series AFA Ultrasonicators.
The 96 microTUBE with AFA fiber, are permanently mounted to form an integrated and unitized plate assembly that meets SBS (Society of Biomolecular Screening specifications. The tubes are sealed with a pre-pierced foil to allow for low insertion/extraction force so the 96 microTUBE plate and strip are suitable for manual or automated pipetting systems. Importantly; microTUBE uses a Covaris specified, high-grade, hardened glass for low acoustic impedance and a better transmission of AFA energy without excessive heat generation. In addition, the microTUBE unique geometry provides synergistic acoustic focusing.

The miniTUBE was specifically engineered to shear genomic DNA into selected fragment sizes ranging from 2 to 5kb. Interaction between the AFA Textured Surface and AFA Ultrasonic energy creates controlled non-inertial cavitation which generates micro-streaming in the sample. DNA trapped in this micro-streaming is gently sheared.
The Clear, the Blue, and the Red miniTUBE each have a different AFA textured surfaces and shearing rate. In all cases, the shearing force is gentle to generate large DNA fragment with a tight fragment size distribution, yet is strong enough to ensure complete sample processing.
