Integrated Microscale Analysis System for Targeted Liquid Chromatography Mass Spectrometry Proteomics on Limited Amounts of Enriched Cell Populations

A complete integrated streamlined microscale workflow is described that is able to accept limited amounts of whole cells from existing enrichment technologies such as FACS (fluorescent activated cell sorter) in a microfluidic device of <5 μL and then prepare and analyze their contents by proteomic nanoLCMS in an online fashion. The FACS-Chip-LCMS workflow was demonstrated by analyzing 5000 enriched cells from a heterogeneous population by LCMS and its performance was assessed by its ability to quantitate 17 proteins ranging more than 4 orders of magnitude.
